| HGNC Family | Non-coding RNAs |
| Name | microRNA 514a-1 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐514a-3p (encoded by MIR514A1) has been identified as a melanoma-specific miRNA that is markedly overexpressed in melanoma compared to other solid cancers. It forms part of the miR-506–514 cluster and has been implicated in the initiation of melanocyte transformation and promotion of melanoma growth. Functional studies reveal that miR‐514a-3p modulates the expression of numerous genes—including CTNNB1, CDK2, MC1R, and notably NF1—thereby contributing to the regulation of key signaling pathways. Direct targeting of NF1 by miR‐514a-3p, as confirmed by luciferase-reporter assays, results in decreased NF1 levels; this downregulation is associated with enhanced survival of BRAFV600E melanoma cells treated with targeted inhibitors, providing a mechanistic insight into drug resistance in melanoma."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in melanoma, miR‐514a-3p exerts significant function in testicular germ cell tumors. In these tumors, overexpression of miR‐514a-3p induces apoptosis through the direct suppression of the paternally expressed gene 3 (PEG3). Loss or downregulation of miR‐514a-3p leads to increased PEG3 expression, which in turn recruits TRAF2 and activates the NF‑κB pathway, ultimately protecting tumor cells from apoptosis. These findings underscore a context-dependent role for MIR514A1, where its regulatory impact on PEG3 and NF‑κB signaling balances cell survival and death, thereby contributing to tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Mitchell S Stark, Vanessa F Bonazzi, Glen M Boyle, et al.
