MIR514A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 514a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn melanoma, miR-514a-3p (MIR514A2) is a key member of the miR-506-514 cluster that is highly enriched in melanoma cells relative to other solid tumors. Its expression is associated with the initiation of melanocyte transformation and melanoma growth. Functionally, miR-514a-3p directly targets several genes, including NF1, a critical regulator of the MAPK pathway; by repressing NF1, miR-514a-3p facilitates increased cell survival, particularly in the context of BRAFV600E mutation and treatment with BRAF inhibitors, thereby contributing to therapeutic resistance."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nIn testicular germ cell tumor (TGCT), miR-514a-3p assumes a contrasting role by promoting apoptosis. It achieves this by directly downregulating PEG3, a gene that, when overexpressed, recruits TRAF2 and activates the NF-κB pathway, thereby protecting germ cells from apoptosis. Loss of miR-514a-3p expression in TGCT leads to elevated PEG3 levels, increased nuclear accumulation of NF-κB subunits, and enhanced survival signaling, highlighting the importance of tightly regulated miR-514a-3p levels in maintaining the balance between cell survival and programmed cell death"}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Mitchell S Stark, Vanessa F Bonazzi, Glen M Boyle, et al.
