MIR515-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 515-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‐515, predominantly the 5p species (and in one report the 3p isoform), has emerged as a critical tumor‐suppressive microRNA in several cancer types. In breast, lung, and liver cancers, its expression is frequently down‐regulated, thereby relieving repression of key oncogenic mediators. For instance, miR‐515‐5p directly targets MARK4 to inhibit cancer cell migration and metastasis, while it also counteracts estrogen‐driven proliferation in breast cancer by repressing sphingosine kinase 1 (SK1) activity. Moreover, in non‐small cell lung cancer (NSCLC), miR‐515‐5p reduces cell survival and metastasis through direct targeting of chemokine CXCL6, and in hepatocellular carcinoma its overexpression impairs the IL‐6/JAK/STAT3 cascade to inhibit invasion."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": "Disruptions in miR‐515 regulation can occur not only at the transcriptional level but also through competitive endogenous RNA mechanisms and genetic variations. A single nucleotide polymorphism in the 3′‐UTR of IGF‐1R that interferes with miR‐515‐5p binding has been associated with increased IGF‐1R levels and modified breast cancer risk in BRCA1 mutation carriers. Moreover, several oncogenic circular RNAs (circRNAs) and long non‐coding RNAs (lncRNAs) have been shown to “sponge” miR‐515‐5p in various malignancies including gastric, prostate, oral squamous cell, and cutaneous squamous cell carcinomas. These sponging events relieve miR‐515‐mediated suppression of downstream targets such as XIAP, TGF‐β1, FKBP1A, TPX2, and SLIT3. In addition, separate investigations into gastric cancer have examined the related miR‐515‐3p, reinforcing the concept that disruption of miR‐515 activity contributes to tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "12"}]}, {"type": "t", "text": "Beyond its anticancer functions, miR‐515‐5p contributes to the regulation of cell cycle progression and non‐tumorigenic pathophysiological processes. For example, in colorectal cancer, exogenous miR‐515‐5p induces G1 phase arrest and promotes apoptosis by targeting MAD2L1. In noncancer settings, elevated circulating miR‐515‐5p in maternal serum appears to impair trophoblast proliferation and invasion by suppressing XIAP, while in osteoarthritis models its overexpression attenuates chondrocyte apoptosis and the inflammatory response."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "13", "end_ref": "15"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Filipa G Pinho, Adam E Frampton, Joao Nunes, et al.
