| HGNC Family | Non-coding RNAs |
| Name | microRNA 515-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In non‐small cell lung cancer, for example, CXCL6 reduces miR‐515‐5p expression, thereby relieving its tumor‐suppressive effects on survival and metastasis. Similarly, in hepatocellular carcinoma, miR‐515‐5p is substantially downregulated; its overexpression directly targets the 3′‐UTR of IL6 and consequently suppresses the IL‐6/JAK/STAT3 signaling cascade, leading to decreased cell migration and invasion."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition, miR‐515‐5p plays important roles in other malignancies. In breast cancer, disruption of its binding to the igf‐1r 3′‐UTR by a single nucleotide polymorphism results in increased IGF‐1R protein levels, which correlates with earlier disease onset. In colorectal cancer, miR‐515‐5p directly targets MAD2L1, and its overexpression leads to cell cycle arrest at the G1 phase and induces apoptosis. Moreover, members of the miR‐515 family (including miR‐515‐3p) have been implicated in gastric cancer development, underscoring the broad impact of these microRNAs on oncogenic pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nRegulatory mechanisms affecting miR‐515‐5p activity also include sequestration by circular RNAs. In prostate cancer, circ‐PAPPA acts as a sponge for miR‐515‐5p, leading to elevated expression of FKBP1A and enhanced malignant phenotypes. Similarly, in cutaneous squamous cell carcinoma, circ‐LARP1B sequesters miR‐515‐5p, which results in the upregulation of TPX2, thereby promoting cell proliferation, migration, and glycolytic metabolism."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond these oncological settings, miR‐515‐5p also mediates important functions in other disease states. In the context of preeclampsia, miR‐515‐5p is found at elevated levels in maternal serum, and its overexpression in trophoblast cells suppresses XIAP expression, thereby inhibiting cell proliferation and invasion.
