MIR516B2 Gene

HGNC Family Non-coding RNAs
Name microRNA 516b-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent findings indicate that MIR516B2, through its mature miRNA forms (miR‐516b‐5p), functions predominantly as a tumor suppressor in various cancer types. In esophageal squamous cell carcinoma, for example, miR‐516b has been suggested to exert tumor suppressive effects, although its precise regulatory targets in this context remain to be fully defined."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn non‐small cell lung cancer, dysregulation of competing endogenous RNA networks has been implicated in disease progression. Here, the long noncoding RNA FEZF1‐AS1 was found to be upregulated and to act as a molecular sponge for miR‐516b, leading to the de‐repression of ITGA11, a gene whose increased expression is associated with enhanced cell proliferation and migration. A similar ceRNA mechanism was observed in thyroid cancer, where ARAP1‐AS1 sequesters miR‐516b‐5p to relieve its inhibitory effect on PDE5A, thereby promoting cell growth and survival. These observations underscore a critical role for MIR516B2 in modulating oncogenic pathways through its interactions with specific long noncoding RNAs."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, studies in glioblastoma cell models have revealed that enforced overexpression of miR‐516b‐5p can significantly impair cellular migration and invasion, implicating this miRNA in the regulation of cell adhesion and junction dynamics—processes that are critical for tumor metastasis. Although proliferation rates in these models were not affected, the inhibition of invasive behavior supports a broader antimetastatic role for MIR516B2 in central nervous system malignancies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yufeng Zhao, Yaqiang Wang, Guochen Xing "}, {"type": "b", "children": [{"type": "t", "text": "miR-516b functions as a tumor suppressor by directly modulating CCNG1 expression in esophageal squamous cell carcinoma."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Biomed Pharmacother (2018)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1016/j.biopha.2018.07.074"}], "href": "https://doi.org/10.1016/j.biopha.2018.07.074"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "30119241"}], "href": "https://pubmed.ncbi.nlm.nih.gov/30119241"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Heng Song, Hui Li, Xiaosong Ding, et al.
