| Name | microRNA 517b |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA MIR517B, a member of a placenta‐specific miRNA cluster, has been implicated in the regulation of placental function. In preeclamptic pregnancies, increased expression of MIR517B (along with related miRNAs such as miR‑517a and miR‑517c) has been observed in both term and preterm cases. Experimental studies using first trimester extravillous trophoblasts demonstrated that hypoxic conditions induce MIR517B expression, which in turn reduces trophoblast invasion and upregulates anti‑angiogenic factors like sFLT1 via the cytokine TNFSF15. These findings suggest that aberrant MIR517B activity may contribute to the placental dysfunction underlying preeclampsia."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in preeclampsia, MIR517B has been identified among a set of placenta‑associated miRNAs whose expression profiles are altered in conditions such as complete hydatidiform mole, hinting at its broader involvement in placental pathologies. Another study specifically targeting the role of MIR517‐5p in preeclampsia further underscores the potential mechanistic contributions of this miRNA in mediating placental responses to pathological stress."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond placental biology, recent investigations have extended the relevance of MIR517B to oncological contexts. In hepatocellular carcinoma, particularly in patients with portal vein tumor thrombus, altered expression patterns of MIR517B isoforms (e.g., miR‑517b‑3p) have been associated with disease progression and poorer prognosis. These preliminary observations suggest that MIR517B could serve as a prognostic biomarker and may actively participate in tumor biology through the modulation of cellular invasion and metastasis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Lauren Anton, Anthony O Olarerin-George, John B Hogenesch, et al.
