MIR517C Gene

HGNC Family Non-coding RNAs
Name microRNA 517c
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In primary central nervous system primitive neuroectodermal tumors, MIR517C is strikingly overexpressed due to a high‐level amplicon and acts as an oncogene by promoting cell survival, proliferation, and anchorage‐independent growth of human neural stem cells through mechanisms that include upregulation of the WNT pathway and inhibition of differentiation. In parallel, studies in hepatocellular carcinoma have shown that ectopic expression of MIR517C impedes cell proliferation by enforcing a G2/M cell cycle arrest via targeting of proteins such as Pyk2, with its downregulation contributing to tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Disparate yet critical functions of MIR517C are also evident in the regulation of cellular invasion and stress responses. In placental trophoblasts, MIR517C expression is induced under hypoxic conditions and is implicated in the pathogenesis of preeclampsia by reducing trophoblast invasion and enhancing the expression of anti-angiogenic factors. Similarly, in glioblastoma cells harboring wild-type TP53, MIR517C suppresses autophagy and cellular migration by degrading KPNA2 and disrupting the nuclear translocation of TP53, thereby preventing the epithelial-to-mesenchymal transition and correlating with improved patient prognosis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Meihua Li, Kyle F Lee, Yuntao Lu, et al.
