MIR518A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 518a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn imatinib mesylate‐resistant gastrointestinal stromal tumors, miR-518a-5p has been shown to suppress tumor cell proliferation and promote apoptosis through direct targeting of the 3′ untranslated region of the gene PIK3C2A. Reduced levels of this miRNA were found to correlate with increased expression of PIK3C2A, which likely contributes to the cellular resistance observed during imatinib treatment. These findings highlight the importance of miR-518a-5p in modulating drug response in GISTs."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in GISTs, miR-518a-5p has been implicated in the regulation of the chemokine receptor CCR6, a key player in colorectal cancer progression and diffuse large B cell lymphoma (DLBCL). Functional studies demonstrate that miR-518a-5p negatively regulates CCR6 expression, thereby inhibiting cell proliferation and invasion. In DLBCL, this regulatory relationship is further influenced by epigenetic modifications such as promoter hypermethylation and altered histone acetylation patterns. The modulation of downstream JAK2/STAT6 signaling by these miRNA-CCR6 interactions provides further insight into their roles in tumorigenesis and suggests a feedback loop that contributes to lymphoma development."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nEmerging evidence also underscores the potential significance of miR-518a-5p in other malignancies such as breast cancer, where unraveling its molecular interactions may lead to the identification of novel therapeutic targets. As our understanding of miR-518a-5p’s impact on oncogenic pathways deepens, it may become a critical component in the development of strategies to overcome drug resistance and prevent tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yuan Shi, Xiaodong Gao, Qin Hu, et al.
