| HGNC Family | Non-coding RNAs |
| Name | microRNA 520e |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐520e (miR-520e) emerges as a critical regulatory molecule with tumor‐suppressive functions in various human cancers. In hepatocellular carcinoma, miR-520e is markedly downregulated—often via upstream DNA hypermethylation—thereby releasing its direct target, NF-κB-inducing kinase (NIK), which in turn activates downstream signaling (e.g., p-ERK1/2/NF-κB) that promotes hepatoma cell proliferation. These findings underscore miR-520e’s role in curbing tumor growth through inhibition of oncogenic signaling pathways."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its function in liver cancer, miR-520e has been implicated in other pathological states. In visceral adipose tissue, it partners with the regulation of RAB11A—an important effector in glucose homeostasis and lipid metabolism—suggesting that miR-520e may contribute to metabolic disturbances in obesity. In gastric cancer, it is identified as part of the miR-520b/e group displaying tumor-suppressive activity, potentially linked to modulation of epidermal growth factor receptor (EGFR) signaling. Furthermore, in non-small-cell lung cancer (NSCLC), miR-520e suppresses malignant progression by targeting the transcription factor Zbtb7a and influencing Wnt signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "2", "end_ref": "4"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nmiR-520e also plays a notable role in virus-associated and other cancers. In hepatitis B virus (HBV)-positive hepatocellular carcinoma, miR-520e modulates EphA2 expression and, by extension, disrupts p38MAPK/ERK1/2 signaling to simultaneously reduce viral replication and tumor growth. In gliomas, overexpression of miR-520e restricts cell proliferation and invasion by downregulating FGF19 and subsequent Wnt/β-catenin signaling. Finally, in osteosarcoma, the regulatory interplay between long noncoding RNA ZFAS1 and miR-520e affects RHOC expression, thereby influencing tumor progression. These studies collectively highlight the multifaceted role of miR-520e as a tumor suppressor and a potential therapeutic target in diverse pathological contexts."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "7"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "S Zhang, C Shan, G Kong, et al.
