MIR521-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 521-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSperm lysozyme‐like protein 1 (SLLP1) is a specialized acrosomal protein that, despite its structural similarity to conventional c‐lysozymes, lacks bacteriolytic activity and plays a pivotal role during fertilization. SLLP1 is retained in the equatorial segment of acrosome-reacted sperm and promotes the binding of spermatozoa to the egg plasma membrane. Functional studies using recombinant SLLP1 and its inhibitory antibodies have shown that interference with SLLP1 significantly reduces sperm–oocyte binding and subsequent fertilization, underscoring its essential role in gamete recognition and fusion."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nFurther elucidation of the molecular basis of fertilization revealed that SLLP1 interacts with a unique oocyte receptor, SAS1B—a membrane-associated metalloproteinase expressed exclusively in ovarian tissues. SAS1B localizes to the microvillar domain of oocytes where it binds SLLP1 with high affinity; disruption of this interaction, whether by protease inhibition or genetic knockout, results in impaired fertilization. These observations highlight the SLLP1–SAS1B pairing as a critical mediator of sperm–oocyte adhesion and fusion."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nComplementary structural and biochemical analyses further reveal that SLLP1 exhibits distinct modifications compared with conventional lysozymes, including substitutions at key catalytic residues and the presence of invariant surface “fingerprint” regions. These features, along with the formation of oligomeric helical assemblies, are thought to be important for its selective binding to SAS1B. Moreover, studies in bovine and murine models—as well as observations of autoantibody reactivity in infertile women—suggest that SLLP1 not only plays a role in sperm function but may also be involved in female fertility. This multifaceted role reinforces the significance of SLLP1 in ensuring successful gamete interaction and embryo viability."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "María Belén Herrero, Arabinda Mandal, Laura C Digilio, et al.
