| HGNC Family | Non-coding RNAs |
| Name | microRNA 532 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR532, encompassing its isoforms miR‑532‑3p and miR‑532‑5p, has emerged as a multifunctional regulator in diverse cancers. In colorectal cancer, for example, MIR532‑3p is involved in circRNA‐mediated networks that modulate FOXO4 expression and act to restrain cell growth, metastasis, and epithelial–mesenchymal transition via direct targeting of ETS1 and TGM2. In hepatocellular carcinoma, decreased miR‑532‑3p levels lead to upregulation of oncogenic factors such as KIFC1, thereby promoting metastasis; similar tumor‐suppressive functions are observed in lung adenocarcinoma where miR‑532‑5p directly targets KRAS and MKL2. Conversely, in gastric cancer miR‑532 is overexpressed and facilitates cell migration and invasion through inhibition of NKD1 and activation of the Wnt/β‑catenin pathway. In several gynecologic and urologic malignancies—including ovarian and bladder cancers as well as clear cell renal cell carcinoma—downregulation of MIR532 correlates with enhanced proliferation, invasion, and poorer patient outcomes; in ovarian cancer, for instance, reduced miR‑532 expression relieves repression of hTERT, while in ccRCC miR‑532‑3p targets TROAP. Notably, a contrasting oncogenic role has been delineated in t(4;14) multiple myeloma, where MIR532 is upregulated and promotes proliferation and metastasis by targeting CAMK2N1."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumorigenesis, MIR532 displays significant promise as a biomarker and functional mediator in several non‐malignant disorders. In prostate cancer, elevated levels of MIR532‑5p in urinary exosomes have been linked to biochemical recurrence. Similarly, increased serum expression of MIR532 is observed in deep vein thrombosis. In rheumatoid arthritis, differential expression of this microRNA—shaped in part by sex‐specific epigenetic regulation—may contribute to disease heterogeneity. MIR532‑5p also participates in the host antiviral response during West Nile virus infection by targeting key genes such as SESTD1 and TAB3, and in cerebral ischemia–reperfusion injury its overexpression attenuates apoptosis, oxidative stress, and inflammation through direct binding to the 3′‑UTR of CXCL1. Furthermore, altered MIR532 expression has been noted in conditions such as sarcopenia, intervertebral disc degeneration, and migraine, underscoring its emerging utility as both a prognostic marker and a potential therapeutic target."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "16", "end_ref": "24"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMolecularly, MIR532 exerts its diverse functions by directly binding to the 3′‑UTRs of target mRNAs across different cellular contexts. It modulates distinct signaling pathways by repressing genes such as FOXO4, ETS1, TGM2, KIFC1, HMGB3, KRAS, MKL2, TROAP, CAMK2N1, CXCL2, and CXCL1. In parallel, epigenetic mechanisms—including promoter hypermethylation and regulation via competing endogenous RNA networks (for example, the MIAT/miR‑532‑3p/STC1 axis)—further fine-tune its expression. In bone and inflammatory disorders, elevated MIR532‑3p levels have been implicated in impairing osteogenic differentiation by downregulating SIRT1 and in modulating pro‑inflammatory signaling; indeed, in macrophages, MIR532‑3p attenuates ASK1/p38 MAPK activation and consequent cytokine release. These findings collectively illuminate a context‑dependent role for MIR532 as either a tumor suppressor or an oncogene, and as a key modulator of inflammation and tissue remodeling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "25", "end_ref": "27"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Longjun Bian, Xiaofei Zhi, Lilin Ma, et al.
