MIR548AH Gene

HGNC Family Non-coding RNAs
Name microRNA 548ah
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

One study documented that ADAM3, a sperm membrane protein, is essential for enabling sperm migration into the oviduct and for the initial binding to the zona pellucida. Its proper folding and cell surface localization depend on a dedicated endoplasmic reticulum chaperone system involving the testis-specific protein disulfide isomerase homolog PDILT and the related chaperone CALR3. In Pdilt(–/–) mice, the absence of this complex led to misfolded ADAM3 and consequent male infertility, despite the sperm’s retained ability to fertilize eggs when surrounded by cumulus cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nA complementary investigation focused on PDILT, a unique member of the protein disulfide isomerase family with an atypical SXXC motif. PDILT is exclusively expressed in the testis and operates as a glycosylated ER chaperone that forms disulfide-dependent complexes with partner proteins, including binding to the oxidoreductase Ero1α. This specialization underlines its non‐redundant role in the precise protein folding and redox regulation needed for the maturation of sperm-specific proteins, thereby ensuring the functional competence of spermatozoa."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAlthough neither abstract explicitly investigates or mentions MIR548AH, its reference in the inquiry suggests an interest in understanding potential regulatory layers within such specialized testicular control mechanisms. Given the established importance of protein folding machineries in sperm function, future studies addressing MIR548AH might explore whether this microRNA modulates the expression or function of key chaperones like PDILT or substrates such as ADAM3. Such insights could uncover a novel facet of post‐transcriptional regulation in testis-specific protein networks."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Keizo Tokuhiro, Masahito Ikawa, Adam M Benham, et al.
