MIR548AP Gene

Name microRNA 548ap
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR548AP, also known as cystatin 10 or carminerin (encoded by Cst10), is a cartilage‐specific molecule that plays a critical role during chondrocyte maturation. Its expression is upregulated as chondrocytes progress from the prehypertrophic to hypertrophic stage, where it facilitates the calcification process essential to endochondral ossification. Notably, MIR548AP contributes to both normal skeletal development and pathologic conditions by transcriptionally inhibiting the expression of key regulators such as nucleotide pyrophosphatase phosphodiesterase 1 (NPP1), thereby modulating chondrocyte mineralization."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n\nDisruption of MIR548AP expression—as demonstrated in knockout models—leads to a reduction in the calcification of hypertrophic chondrocytes. This deficiency manifests not only during physiological growth but also under pathological conditions, such as impaired formation of osteoarthritic osteophytes, ectopic ossification with aging, and delayed healing of bone fractures. The modulation of NPP1 activity appears to be a key mechanism underlying these effects, highlighting MIR548AP’s role as a pivotal regulator in the later stages of endochondral ossification."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n\nIn parallel, in vitro overexpression studies using chondrogenic cells have revealed that elevated levels of MIR548AP accelerate hypertrophic differentiation. This is evidenced by an earlier appearance of type X collagen, increased mineral deposition seen by stronger alizarin red staining, and the activation of apoptotic markers such as caspase activity and annexin V expression. Together, these findings support a dual role for MIR548AP in promoting both terminal chondrocyte maturation and programmed cell death, making it a critical mediator in cartilage development and potential target in disorders of pathological ossification."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yu Koshizuka, Takashi Yamada, Kazuto Hoshi, et al.
