MIR548B Gene

HGNC Family Non-coding RNAs
Name microRNA 548b
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn non‐small cell lung cancer, miR‑548b has been shown to be modulated by upstream regulators such as Axl kinase, contributing to gefitinib resistance by promoting cell cycle arrest, enhancing gefitinib‑induced apoptosis, and reducing epithelial‐to‐mesenchymal transition, migration, and tumorigenesis via targeting key genes (e.g., CCNB1). In addition, genetic variants (regQTL‑SNPs) can influence the expression pattern of miR‑548b‐3p and its interplay with genes like LAMC1, thereby modulating lung cancer risk, while expression changes of miRNAs—including miR‑548b—have also been monitored in models of lung ischemia‑reperfusion injury."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn gliomas and melanoma, miR‑548b behaves chiefly as a tumor suppressor. In glioma cells its underexpression is linked to enhanced proliferation and invasiveness, and forced re‑expression leads to growth inhibition and reduced colony formation primarily through direct targeting of metastasis‑associated proteins such as MTA2. Similarly, in melanoma, diminished levels of miR‑548b correlate with poor overall survival, whereas its overexpression reduces tumor growth and metastatic traits by directly downregulating HMGB1. Moreover, in colorectal cancer, miR‑548 (encompassing related isoforms) is implicated in regulating tumor development, notably by modulating pathways involving WNT2 signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "6"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn breast and gastric cancers, the tumor‐suppressive potential of miR‑548b is further underscored. In breast cancer, studies are characterizing the expression and biological functions of miR‑548b‑3p, suggesting that its deregulation may be involved in disease progression. In gastric carcinoma cells, miR‑548b‑5p has been demonstrated to inhibit cell viability, migration, and invasion by directly targeting frizzled 7 (FZD7), thereby promoting apoptosis through modulation of apoptotic regulators. Collectively, these findings across multiple cancer types indicate that restoring miR‑548b expression might offer therapeutic benefits by curtailing malignant progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "9"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Y Wang, H Xia, Z Zhuang, et al.
