MIR548C Gene

HGNC Family Non-coding RNAs
Name microRNA 548c
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐548c is emerging as an important regulatory molecule in diverse physiological and pathological contexts. In the cardiovascular system, profiling of peripheral blood mononuclear cells from patients with dilated cardiomyopathy and chronic heart failure has revealed a marked downregulation of miR‐548 family members – including miR‐548c – pointing to its potential as an early diagnostic biomarker. In preeclampsia, reduced levels of miR‐548c‐5p in serum exosomes and placental mononuclear cells lead to derepression of its target PTPRO, thereby enhancing macrophage activation via the NF‐κB pathway and contributing to the pronounced inflammatory state observed in the disease."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn oncology, miR‐548c exhibits predominantly tumor‐suppressive functions across a spectrum of malignancies. Investigations in gynecological cancers have highlighted its role in endometrial and ovarian tumorigenesis, suggesting that miR‐548c may serve as a “universe” miRNA in these contexts. In colorectal cancer, for instance, miR‐548c‐5p is significantly downregulated, resulting in elevated levels of the glycolytic enzyme PGK1 and concomitant increases in cellular proliferation and inflammatory cytokine production. Similarly, deregulation of miR‐548c‐3p has been associated with tumor progression in papillary thyroid carcinoma. In addition, studies in breast cancer further underscore the capacity of miR‐548c‐5p to modulate tumor phenotypes, while in hepatocellular carcinoma the loss of miR‐548c‐3p—often through sequestration by oncogenic circular RNAs—leads to overexpression of LAMC2 and promotion of epithelial–mesenchymal transition. In osteosarcoma, miR‐548c‐3p negatively regulates cell proliferation and invasion, further reinforcing its tumor‐suppressive role."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in inflammation and oncogenesis, miR‐548c also plays a part in cellular differentiation processes.
