| HGNC Family | Non-coding RNAs |
| Name | microRNA 548d-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In human bone marrow mesenchymal stem cells, bioinformatic analysis identified miR‐548d‐5p as a candidate regulator of peroxisome proliferator-activated receptor-γ (PPARγ), suggesting that this miRNA may influence the balance between adipogenic and osteogenic differentiation under dexamethasone induction. This observation highlights a potential role of miR‐548d in modulating mesenchymal cell fate and tissue regeneration processes."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of cancer, miR‐548d‐3p has been shown to exert tumor‐suppressive functions. One study demonstrated that miR‐548d‐3p, particularly when co-expressed with miR‐559, specifically interacts with the 3′-UTR of ERBB2 mRNA to cooperatively inhibit its translation in tumors with ERBB2 overexpression. Similarly, reduced levels of miR‐548d‐3p in osteosarcoma correlate with enhanced activity of oncogenic KRAS, with restoration of miR‐548d‐3p markedly reducing cell growth and migration in vitro."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn infectious diseases, particularly leishmaniasis, miR‐548d‐3p appears to play a critical role in modulating the host immune response. In American Tegumentary Leishmaniasis, upregulation of miR‐548d‐3p in infected monocytes and plasma from self-healed patients was associated with altered chemokine production—including MCP1/CCL2, RANTES/CCL5, and IP10/CXCL10—that may favor lesion resolution. Moreover, in visceral leishmaniasis, inhibition of miR‐548d‐3p led to enhanced parasite growth, potentially due to diminished MCP-1 and nitric oxide production, underscoring its significance in orchestrating effective inflammatory responses during infection."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Junkui Sun, Yisheng Wang, Yuebai Li, et al.
