| HGNC Family | Non-coding RNAs |
| Name | microRNA 548i-1 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In primary dendritic cells, exposure to interleukin‐4 during differentiation induces the expression of the adaptor protein RUFY4, which positively regulates macroautophagy via mTORC1 inhibition. RUFY4 enhances the formation of autophagosomes marked by LC3 and Syntaxin 17, thereby promoting lysosome tethering and efficient endogenous antigen presentation that aids in pathogen clearance."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Recent investigations in hepatocellular carcinoma have delineated a distinct regulatory mechanism involving noncoding RNAs. Here, the circular RNA circNFATC3 acts as a molecular sponge for hsa‐miR-548I1, a microRNA that, when active, normally targets the mRNA of NFATC3. By sequestering miR-548I1, circNFATC3 protects NFATC3 from downregulation, leading to inhibition of the phosphorylation cascades of JNK, c-Jun, AKT, and mTOR. This regulatory axis exerts tumor suppressor functions, as restoration of NFATC3 correlates with reduced hepatoma cell proliferation, migration/invasion, and metastasis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "In a separate context, the adaptor protein RUFY4 also plays a critical role in bone remodeling. During osteoclastogenesis, RUFY4 is upregulated and facilitates the fusion between late endosomes and lysosomes by bridging Rab7 on endosomes with LAMP2 on lysosomes, a process essential for the exocytotic release of degradative enzymes such as cathepsin K. Disruption of RUFY4 impairs lysosomal trafficking, leading to defective bone resorption and protection against pathological bone loss in models of osteoclast‐mediated disease."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Seigo Terawaki, Voahirana Camosseto, Francesca Prete, et al.
