MIR548J Gene

HGNC Family Non-coding RNAs
Name microRNA 548j
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent investigations into the regulatory roles of miRNAs have revealed that miR‑548j, a member of the miR‑548 family, may function by interacting directly with the coding regions of target mRNAs. In a study of conserved miRNA binding sites, miR‑548j was identified alongside its homologues as having significant binding sites within the coding sequence of genes such as PTPN12. These binding sites encode specific oligopeptides (e.g. PRTRSC) and are highly conserved among orthologous genes, suggesting that miR‑548j could modulate protein expression via translational repression or alteration of mRNA stability."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a separate clinical and experimental setting, a study focusing on peripheral arterial disease (PAD) found that circulating levels of miR‑548j‑5p were significantly reduced in PAD patients. Functional assays demonstrated that up‑regulation of miR‑548j‑5p enhanced the angiogenic capacity of endothelial progenitor cells by stimulating cell migration and tube formation, likely via modulation of endothelial nitric oxide synthase (NOS) and stromal cell‑derived factor‑1 (SDF‑1) signaling pathways. In vivo, restoration of miR‑548j‑5p improved capillary density and perfusion recovery in ischemic limbs, highlighting its potential therapeutic value for ischemic vascular conditions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Anatoliy T Ivashchenko, Assel S Issabekova, Olga A Berillo "}, {"type": "b", "children": [{"type": "t", "text": "miR-1279, miR-548j, miR-548m, and miR-548d-5p binding sites in CDSs of paralogous and orthologous PTPN12, MSH6, and ZEB1 Genes."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Biomed Res Int (2013)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1155/2013/902467"}], "href": "https://doi.org/10.1155/2013/902467"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "23957009"}], "href": "https://pubmed.ncbi.nlm.nih.gov/23957009"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Chiu-Yang Lee, Shing-Jong Lin, Tao-Cheng Wu "}, {"type": "b", "children": [{"type": "t", "text": "miR-548j-5p regulates angiogenesis in peripheral artery disease."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Sci Rep (2022)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1038/s41598-022-04770-6"}], "href": "https://doi.org/10.1038/s41598-022-04770-6"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "35039547"}], "href": "https://pubmed.ncbi.nlm.nih.gov/35039547"}]}]}]}
