MIR550A1 Gene

HGNC Family Non-coding RNAs
Name microRNA 550a-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMembers of the miR-550 family, including miR-550a-3p and miR-550-1, have emerged as important regulators in diverse biological contexts. In breast cancer, for example, miR-550a-3p expression is inversely correlated with the levels of key Ras/ERK pathway effectors (ERK1 and ERK2), and its gradual downregulation during tumor progression has been associated with poorer overall and disease‐free survival. In parallel, studies in diabetic patients have revealed that in peripheral B cells, TNF‑α stimulation elevates miR-550a levels, which in turn mediates chromatin remodeling at the cyp27b1 gene locus, impairing the conversion of vitamin D metabolites and contributing to reduced bone mineral density."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCirculating levels of miR-550a-3p are also emerging as pertinent to bone health. In patients with idiopathic osteoporosis, serum miR-550a-3p levels correlate with parameters of dynamic bone formation and with detailed microarchitectural features, suggesting a potential role as a surrogate biomarker for assessing bone quality. In primary melanoma, reduced expression of miR-550 has been identified and is proposed to hold prognostic value, further underlining the multifaceted roles of this miRNA family in tumor biology."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn hematologic malignancies and cancer stem cell biology, the miR-550 family also demonstrates tumor-suppressive properties. In acute myeloid leukemia, enforced expression of miR-550-1 impairs cell proliferation by targeting the WWTR1 gene and by downregulating the m6A methyltransferase cofactor WTAP, thereby destabilizing oncogenic transcripts. Additionally, within breast cancer stem cells, the LINC00261/miR-550a-3p/SDPR regulatory axis has been shown to affect cell viability, microsphere formation, migration, and invasion—where LINC00261 functions as a sponge for miR-550a-3p, resulting in upregulation of SDPR and consequent suppression of malignant stem cell traits."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jar-Yi Ho, Ren-Jun Hsu, Chih-Hsi Wu, et al.
