| HGNC Family | Non-coding RNAs |
| Name | microRNA 551b |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have revealed that miR‐551b plays multifaceted, context‐dependent roles in human diseases. In several tumor types, including ovarian cancer, triple‐negative breast cancer, lung cancer, and papillary thyroid cancer, upregulation of specific miR‐551b isoforms (miR‐551b‐3p/‐5p) drives oncogenic processes. For example, in ovarian cancer stem cells, enhanced miR‐551b expression promotes proliferation, invasion, and chemoresistance by suppressing key tumor suppressors such as Foxo3 and TRIM31, while in triple‐negative breast cancer, genomic amplification leads to increased miR‐551b‐3p that translocates to the nucleus to activate STAT3‐mediated oncostatin signaling. In chemoresistant lung cancer cells, overexpressed miR‐551b contributes to acquired drug resistance by reducing catalase levels and potentiating EGFR‐Akt survival signaling, and in papillary thyroid cancer, high miR‐551b levels are associated with increased malignancy through inhibition of ERBB4."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nConversely, in several gastrointestinal and related malignancies miR‐551b appears to act as a tumor suppressor. In gastric cancer, diminished expression of miR‐551b correlates with epithelial–mesenchymal transition (EMT), metastasis, and poor prognosis, with its restoration inhibiting malignant behaviors via direct targeting of factors such as ERBB4 and TMPRSS4. Similarly, in cholangiocarcinoma, miR‐551b suppresses cell proliferation and induces apoptosis by downregulating Cyclin D1, while in gallbladder cancer, enforced miR‐551b expression reverses EMT by targeting H6PD. In colorectal cancer, studies have shown that miR‐551b (sequestered by oncogenic circRNAs in some settings) modulates cell cycle progression via CDK6, and exosomal delivery of miR‐551b from bone marrow–derived stromal cells can inhibit breast cancer cell growth and migration. Moreover, circulating levels of miR‐551b may serve as promising diagnostic biomarkers in gastric cancer."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its direct roles in tumor biology, aberrant circulating levels of miR‐551b have been linked to inflammatory and premalignant conditions.
