MIR557 Gene

HGNC Family Non-coding RNAs
Name microRNA 557
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have revealed a tumor‐suppressive role for miR-557 in lung cancer. Reduced expression of miR-557 was observed in clinical lung cancer specimens and cell lines, where its overexpression decreased cell viability and invasion while promoting cell death through downregulation of lymphocyte enhancement factor 1 (via its 3' untranslated region). These findings indicate that miR-557 acts to inhibit lung cancer cell proliferation and invasion, suggesting its potential as a tumor suppressor."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "MiR-557 also appears to play a significant role in immune thrombocytopenia (ITP). In experimental ITP models, enhanced miR-557 expression was associated with impaired proliferation and maturation of thrombopoietin-induced megakaryocytes, along with increased apoptotic signaling. Conversely, inhibition of miR-557 improved the physiological function of megakaryocytes, increased platelet counts, modulated apoptosis-related gene expression, and activated the Akt/ERK signaling pathway, collectively ameliorating ITP symptoms."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "Moreover, in hepatocellular carcinoma (HCC), miR-557 is down-regulated in tumor tissues and cell lines and is linked to tumor recurrence and metastasis. Restoration of miR-557 levels inhibited HCC cell proliferation, migration, invasion, and epithelial-to-mesenchymal transition by directly targeting RAB10 and suppressing the Wnt/β-catenin signaling cascade. This evidence reinforces the concept that miR-557 functions as a tumor suppressor across diverse cancer types by modulating key oncogenic pathways."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jiayong Qiu, Yingying Hao, Shenshen Huang, et al.
