MIR561 Gene

HGNC Family Non-coding RNAs
Name microRNA 561
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR-561 and its isoform miR-561-5p have emerged as pivotal regulators in multiple cancers. In hepatocellular carcinoma, miR-561-5p is markedly overexpressed in metastatic tissues where it promotes lung metastasis by directly targeting CX3CL1, thereby impairing the chemotactic recruitment and cytotoxic function of CX3CR1+ natural killer cells. In non‐small cell lung cancer, however, miR-561 functions as a tumor suppressor by downregulating P-REX2a, which leads to cell cycle arrest, inhibition of cell proliferation, and induction of apoptosis. In pancreatic ductal adenocarcinoma, miR-561-5p has been implicated in the regulation of RAC1 expression, affecting cellular proliferation, migration, invasion, and apoptosis. Coincidentally, a functional variant in the TYMS gene—predicted to alter miR-561 binding—has been associated with an increased risk of breast cancer, underscoring the influence of miR-561 on gene expression modulation in tumorigenesis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in cancer, miR-561 is also a critical mediator in inflammatory and neuronal processes. In the inflammatory context of gouty arthritis, the circular RNA circHIPK3 sequesters miR-561, thereby relieving its suppression of key inflammatory mediators TLR4 and NLRP3 and promoting increased cytokine production. Similarly, during neuronal differentiation, the circular RNA hsa_circ_0002468 acts as a molecular sponge for miR-561, which in turn modulates its target E2F8; this interaction helps balance the proliferative and differentiative capacities of neuronal cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn multiple myeloma, the tumor‐suppressive long non-coding RNA NBR2 exerts its function by modulating the miR-561-5p/DLC1 axis. NBR2 downregulates miR-561-5p, thereby alleviating its repression on DLC1, which results in the activation of the AMPK/mTOR signaling pathway, attenuation of glycolysis, and reduction of cell viability.
