MIR564 Gene

HGNC Family Non-coding RNAs
Name microRNA 564
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have revealed that microRNA‐564 (miR-564) functions as a potent tumor suppressor in multiple cancer types. In breast cancer cells, overexpression of miR-564 concurrently inhibits both the PI3K and MAPK signaling cascades by directly targeting oncogenic effectors—including AKT2, GNA12, GYS1, and SRF—thereby enforcing G1 cell cycle arrest, blocking epithelial–mesenchymal transition, and reducing migratory and invasive capacities. In a similar manner, restoration of miR-564 levels in glioblastoma cells reduces TGF-β1 expression and downstream effectors such as phosphorylated SMAD proteins, EGFR, and MMP9, which culminates in a marked decrease in cell proliferation and tumor growth in vivo."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional evidence from studies in lung and bone malignancies further underscores the tumor-suppressive role of miR-564. In lung cancer, low miR-564 expression is significantly associated with poorer overall survival, suggesting its potential as a prognostic marker. In osteosarcoma, forced expression of miR-564 directly targets Akt, which results in diminished glycolytic metabolism, suppression of cell proliferation, and induction of apoptosis; clinical investigations have begun to assess its significance in patient outcomes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nEmerging data also indicate that miR-564 may have roles beyond traditional tumor suppression. Preliminary studies in hypertrophic scar models propose that miR-564 could modulate fibrotic or proliferative responses in scar formation, while investigations in esophageal squamous cell carcinoma have demonstrated that its activity is impaired by sponging via a circular RNA (circ_0003340). In this context, miR-564 normally targets the 3′ untranslated region of TPX2 to restrict cell proliferation and invasion; reducing circ_0003340 levels or restoring miR-564 function in esophageal cancer cells leads to significant antitumor effects both in vitro and in vivo."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Merve Mutlu, Özge Saatci, Suhail A Ansari, et al.
