| HGNC Family | Non-coding RNAs |
| Name | microRNA 568 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Its expression decreases upon T‐cell activation, and forced over‐expression using miR-568 mimics attenuates key activation markers (CD25, CD69, CD154), diminishes IL-2 production, and limits cell proliferation. In regulatory T cells (Treg), elevated levels of MIR568 also suppress NFAT5 expression, resulting in reduced production of immunosuppressive cytokines (TGF-β and IL-10), impaired Treg differentiation, and weakened suppression of effector cells. These findings indicate that MIR568 modulates both the activation of conventional CD4(+) T cells and the regulatory functions of Treg cells, suggesting its potential as a target for inflammatory and autoimmune disease treatments."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Disruptive inflammation in the lung endothelium associated with conditions such as ARDS and VILI is also subject to posttranscriptional control by MIR568. In human lung endothelial cells, MIR568 binds to the 3′-UTR of the proinflammatory molecule PBEF/NAMPT, thereby attenuating its transcriptional and translational upregulation in response to bioactive agonists like cyclic stretch and LPS. This inhibitory modulation of PBEF/NAMPT by MIR568 suggests a therapeutic avenue for reducing inflammatory lung injury."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Wei Li, Ling-bo Kong, Jun-Tang Li, et al.
