MIR581 Gene

HGNC Family Non-coding RNAs
Name microRNA 581
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In one study, miR-581 was found to promote hepatitis B virus surface antigen (HBsAg) expression by targeting and downregulating both Dicer and EDEM1. This regulation appears to increase HBsAg secretion and enhance cell proliferation, suggesting that altered miR-581 levels during hepatocarcinogenesis could contribute to disease progression by impacting viral antigen expression and cellular growth."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn colorectal cancer, miR-581 has emerged as a regulator of metastasis. It directly targets SMAD7 to modulate the TGFβ signaling pathway, thereby influencing epithelial-to-mesenchymal transition, invasion, and cell survival. Functional assays indicated that inhibition of miR-581 suppresses cell proliferation and induces apoptosis, further underlining its potential role as a driver of metastatic behavior in colorectal tumors."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, in another investigation of hepatocellular carcinoma, miR-581 was implicated in tumor progression through a competing endogenous RNA network. Here, the long non-coding RNA lnc-COX7C-5 serves as a molecular sponge for miR-581, thereby modulating the expression of the downstream target ZEB2. This regulatory axis was associated with enhanced proliferation, migration, invasion, and even lung metastasis in vivo, suggesting that the lnc-COX7C-5/miR-581/ZEB2 pathway may represent a promising therapeutic target in HCC."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yu-Qun Wang, Yong-Feng Ren, Yi-Jiang Song, et al.
