| HGNC Family | Non-coding RNAs |
| Name | microRNA 582 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nResearch on MIR582 – which encompasses both the −3p and −5p mature forms – has revealed context‐dependent roles in cancer. In non–small cell lung carcinoma, increased miR‐582‐3p expression activates the Wnt/β–catenin pathway by concurrently suppressing several negative regulators (AXIN2, DKK3, and SFRP1), thereby promoting cancer stem cell traits and tumor recurrence. In hepatocellular carcinoma, miR‐582–5p is downregulated and its restoration impedes tumor cell proliferation by inhibiting CDK1 and AKT3, while in colorectal cancer conflicting reports indicate that either a loss (via targeting Rab27a) or an increase (via suppression of PTEN) of MIR582 activity—further modulated by circRNAs acting as sponges (for example, hsa_circ_0009361)—may influence tumor progression. In gastric cancer, investigations have dissected miR‐582–5p function on cell growth and, in a related manner, circ_0000423 has been found to promote tumor progression by sponging miR‐582‐3p and upregulating DIXDC1. Additional work in triple–negative breast cancer, ovarian cancer, bladder cancer, melanoma, and acute leukemias (AML and B–cell precursor ALL) consistently suggests that aberrant MIR582 levels alter proliferation, migration/invasion and cell survival via targeting key oncogenic pathways such as NOTCH1, FOXG1, FOXC1, and by affecting mitochondrial metabolism."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncology, MIR582 has been implicated in diverse non–malignant disorders. In the vascular compartment, circulating levels of MIR582 (without clear differentiation of arm origin) have been proposed as promising biomarkers for deep vein thrombosis, while in aortic dissection, reduced miR‐582 expression in smooth muscle cells appears to enhance susceptibility to apoptosis via targeting genes such as NET1. In trophoblasts, miR‐582‐3p directly targets EG–VEGF to downregulate matrix metalloproteinases, thereby restraining invasive capacity—a mechanism relevant to placental development and obstetric complications where maternal plasma levels are aberrant relative to placental expression. Furthermore, a two–base pair deletion in the APP gene 3′UTR that impairs miR‐582–3p binding has been associated with APP overexpression in cerebral amyloid angiopathy. Additionally, in knee osteoarthritis, miR‐582‐5p is implicated in the lineage determination of mesenchymal progenitors via direct targeting of Runx2, and studies in diabetic angiopathy suggest a role in regulating vascular smooth muscle cell repair."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "18", "end_ref": "25"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nRecent studies have additionally underscored the regulation of MIR582 biogenesis and function by non–coding RNAs. For example, certain circular RNAs and long non–coding RNAs (such as RP11–301G19.1) act as sponges to sequester MIR582 (either the 3p or 5p forms), thereby indirectly modulating the expression of downstream targets with roles in antiviral defense (via RIG–I) and multiple myeloma proliferation.
