MIR600 Gene

HGNC Family Non-coding RNAs
Name microRNA 600
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nResearch across several cancer types has demonstrated that miR‑600 plays a pivotal role in modulating tumor cell proliferation and metastatic behavior by suppressing key oncogenic pathways. In cervical cancer models, elevated miR‑600 levels were associated with prolonged patient survival, suggesting a tumor‐suppressive function. In non‑small cell lung cancer, the overexpression of miR‑600—released from sequestration by the oncogenic circRNA circPIP5K1A—suppresses HIF‑1α–mediated signaling, thus inhibiting metastasis and proliferation. Similarly, in HeLa cell studies, miR‑600 was shown to restrain cell proliferation via inhibition of HIF‑1α signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, studies in breast cancer have revealed that reduced expression of miR‑600 is linked to enhanced malignancy and decreased sensitivity to chemotherapeutic agents such as sorafenib. Functional investigations indicate that miR‑600 normally acts to inhibit the expression of EZH2—a negative regulator—thus facilitating the activity of the tumor suppressor RUNX3. Loss of miR‑600 thereby promotes uncontrolled cell proliferation and metastasis, underscoring its role as a tumor suppressor in this context."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nConversely, miR‑600 appears to adopt oncogenic characteristics in ovarian cancer. Here, miR‑600 expression is significantly upregulated in tumor tissues and cancer stem cells, where it directly targets and downregulates the transcription factor KLF9. This interaction enhances cancer cell stemness, proliferation, and metastatic capacities, suggesting that the function of miR‑600 is context‑dependent and may shift from tumor suppression to tumor promotion depending on the cellular environment."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Wang Yong, Xuan Zhuoqi, Wang Baocheng, et al.
