MIR602 Gene

HGNC Family Non-coding RNAs
Name microRNA 602
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Its overexpression correlates with lymph node metastasis, advanced tumor stage and poor overall survival. Mechanistic studies have identified the transcription factor FOXK2 as a direct target of miR‑602; downregulation of FOXK2 by miR‑602 enhances proliferation, metastasis, and cell cycle progression. Furthermore, these diminished levels—alongside elevated levels of S100B—may serve as diagnostic markers, with miR‑602 levels correlating with the severity of neurological deficits. In an in vitro model of oxygen and glucose deprivation/reoxygenation using human brain microvascular endothelial cells, miR‑602 is upregulated and acts protectively by reducing reactive oxygen species production and promoting cell survival. This protective effect is mediated through the activation of the NRF2/ARE pathway, with KEAP1 and HRD1 identified as downstream effectors whose overexpression counteracts miR‑602’s benefits."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn gastric cancer, an interplay between the long noncoding RNA HITT and miR‑602 has been observed. Although patient samples reveal upregulated HITT concomitant with downregulated mature miR‑602, experimental overexpression of HITT in gastric cancer cells specifically increases mature miR‑602 levels without altering its precursor form. Functionally, miR‑602 appears to act as an inhibitor of apoptosis; HITT overexpression further promotes apoptosis in gastric cancer cells by counteracting the anti‑apoptotic effects of miR‑602.
