MIR605 Gene

HGNC Family Non-coding RNAs
Name microRNA 605
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR-605 is a multifunctional microRNA that plays a critical role in regulating the p53 network. Upon cellular stress, wild‐type p53 transcriptionally activates miR-605, which in turn post-transcriptionally represses Mdm2, thereby interrupting the inhibitory p53:Mdm2 feedback loop and enabling rapid p53 accumulation to induce cell cycle arrest and apoptosis. In addition, in Li–Fraumeni syndrome, genetic variants of miR-605 further modulate the phenotype of TP53 mutation carriers, underscoring its importance in tumor suppression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its canonical role in the p53 axis, miR-605 influences tumor behavior in a context-dependent manner by targeting key oncogenic and tumor‐promoting factors. In melanoma, for example, miR-605 suppresses cell growth by negatively regulating INPP4B-mediated SGK3 activation, while in hepatocellular carcinoma its downregulation is associated with enhanced epithelial–mesenchymal transition and metastasis via TRAF6/NF-κB signaling. Similarly, in intrahepatic cholangiocarcinoma miR-605 directly targets PSMD10 to inhibit tumor progression, whereas in non–small cell lung cancer an alternatively processed isoform (miR-605-5p) has been found to promote proliferation and invasion by targeting TNFAIP3."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nmiR-605 functions are further modulated by complex networks involving competitive endogenous RNAs and genetic polymorphisms. In bladder cancer, circ-VANGL1 acts as a sponge for miR-605-3p to derepress VANGL1 and enhance cell migration and invasion, while in glioma, BLACAT1 similarly sequesters miR-605-3p leading to upregulation of VASP and tumor progression. In non–small cell lung cancer, LINC01703 sponges miR-605-3p to upregulate MACC1 and thereby promote aggressiveness. In parallel, multiple studies have investigated single nucleotide polymorphisms in the MIR605 gene (notably rs2043556), reporting associations with susceptibility to gastrointestinal, prostate, and head and neck cancers, as well as with treatment outcomes such as clopidogrel response and the development of multiple primary tumors in TP53 mutation carriers. Moreover, emerging evidence suggests that miR-605 also contributes to the regulation of inflammatory processes, as its downregulation was linked with increased CXCL5 expression and enhanced chemotaxis in complex regional pain syndrome."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jiening Xiao, Huixian Lin, Xiaobin Luo, et al.
