MIR6071 Gene

HGNC Family Non-coding RNAs
Name microRNA 6071
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nThe study demonstrates that miR-6071 acts as a tumor suppressor in hepatocellular carcinoma by regulating liver cancer stem cells (LCSCs). Reduced miR-6071 levels were observed in LCSCs, and its overexpression led to decreased self-renewal, proliferation, migration, and tumorigenic capabilities of HCC cells. Mechanistically, miR-6071 directly targets the 3′UTR of PTPN11 mRNA, resulting in downregulation of this oncogenic phosphatase, as confirmed by bioinformatics analysis and luciferase reporter assays. A significant negative correlation between miR-6071 expression and PTPN11 levels was found in clinical HCC samples, supporting the regulatory link between the two. Functionally, interference with PTPN11, as well as the use of the inhibitor IACS-13909, nullified the effects of miR-6071 modulation on LCSC properties, underscoring the importance of the miR-6071–PTPN11 axis. Furthermore, miR-6071 upregulation enhanced the sensitivity of HCC cells to lenvatinib treatment, and clinical data suggested that higher miR-6071 expression is associated with improved postoperative treatment outcomes."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Minyong Chen, Huaxiang Wang, Songchang Shi, et al.
