| Name | microRNA 6089 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In one investigation, exosome‐encapsulated miR-6089 was found to be significantly decreased in the serum of RA patients compared with healthy controls. Functional experiments revealed that miR-6089 directly targets TLR4 and, by doing so, inhibits lipopolysaccharide-induced proliferation and activation of macrophage-like cells. This downregulation of TLR4 signaling consequently restrains the production of inflammatory cytokines such as IL-6, IL-29, and TNF-α."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a complementary study, researchers demonstrated that the long non-coding RNA HIX003209 functions as a competing endogenous RNA (ceRNA) by sponging miR-6089 in macrophages. This sequestration effectively diminishes the ability of miR-6089 to suppress TLR4/NF-κB pathway activation, thereby exacerbating the inflammatory response. Together, these findings suggest that miR-6089 normally acts as a negative regulator of TLR4-mediated inflammation, and its dysregulation may contribute to the pathogenesis of RA. The dual evidence outlines the therapeutic potential of modulating miR-6089 levels in inflammatory disease states."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Donghua Xu, Meiyan Song, Chunxiang Chai, et al.
