| Name | microRNA 6165 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In human cell lines such as HCT116, HeLa, NT2, SW480, and HEK293T, overexpression of miR-6165 leads to the downregulation of several target genes—including ABLIM-1, PVRL1, PKD1, PLXNA2, and PDK1—which in turn affects neural differentiation, cell cycle progression, and may increase apoptosis. The independent regulation of miR-6165, distinct from its host gene, suggests that it contributes additional complexity to p75NTR-associated cellular functions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn colorectal cancer models, miR-6165 has been shown to exert tumor suppressive effects. Overexpression of miR-6165 in SW480 cells leads to a direct interaction with the 3′UTR of the insulin-like growth factor 1 receptor (IGF-1R) gene, resulting in substantial downregulation of IGF-1R and downstream oncogenic signals—including AKT2, AKT3, PI3KR3, PI3KR5, CCND1, c-MYC, and P21 transcripts—which ultimately promotes apoptosis and reduces cellular viability. Moreover, an inverse expression pattern of miR-6165 and IGF-1R during NT2 cell neural differentiation strengthens the concept of its key regulatory role in both developmental and oncogenic contexts."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditionally, the functional landscape of miR-6165 is further enriched by its interaction with noncoding RNAs in hepatocellular carcinoma. The circular RNA circ_0001459 has been identified as a molecular sponge for miR-6165, thereby mitigating its inhibitory effect on IGF1R expression. This sponging action contributes to the upregulation of IGF1R, leading to enhanced epithelial-to-mesenchymal transition and promoting HCC progression and metastasis.
