MIR617 Gene

HGNC Family Non-coding RNAs
Name microRNA 617
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In psoriatic skin, MIR617 is upregulated in both patient tissues and in interleukin‐22–stimulated keratinocytes, where its increased expression enhances cell proliferation and cell cycle progression while suppressing apoptosis. Mechanistically, MIR617 directly targets FOXO4, leading to its downregulation; overexpression of FOXO4 can counteract these effects, highlighting MIR617’s role in promoting keratinocyte growth and its potential as a therapeutic target in psoriasis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n \nIn contrast, in the context of oral squamous cell carcinoma (OSCC), MIR617 is downregulated compared to normal tissues, a state that contributes to the elevated levels of the oncogenic factor SERPINE1. Experimental evidence demonstrates that restoration of MIR617 levels via mimic transfection reduces SERPINE1 expression, thereby suppressing cell viability and proliferation while promoting apoptosis in OSCC cells. These findings suggest that the MIR617/SERPINE1 axis plays a significant tumor-suppressive role in OSCC and positions MIR617 as a promising target for therapeutic intervention."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Tao Liu, Xiaomei Feng, Yongmei Liao "}, {"type": "b", "children": [{"type": "t", "text": "miR-617 Promotes the Growth of IL-22-Stimulated Keratinocytes Through Regulating FOXO4 Expression."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Biochem Genet (2021)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1007/s10528-020-09997-4"}], "href": "https://doi.org/10.1007/s10528-020-09997-4"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "33211221"}], "href": "https://pubmed.ncbi.nlm.nih.gov/33211221"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Chunguang Zhao, Zhiyun Liu "}, {"type": "b", "children": [{"type": "t", "text": "MicroRNA 617 Targeting SERPINE1 Inhibited the Progression of Oral Squamous Cell Carcinoma."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Mol Cell Biol (2021)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1128/MCB.00565-20"}], "href": "https://doi.org/10.1128/MCB.00565-20"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "33820852"}], "href": "https://pubmed.ncbi.nlm.nih.gov/33820852"}]}]}]}
