MIR626 Gene

HGNC Family Non-coding RNAs
Name microRNA 626
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐626 has emerged as a critical regulator in ophthalmic and neurodegenerative contexts. In retinal pigment epithelium (RPE) and other ophthalmic cells, miR‐626 directly targets the 3′‐UTR of Keap1, thereby downregulating its expression and stabilizing nuclear factor erythroid 2-related factor 2 (Nrf2). Activation of the Nrf2 cascade subsequently promotes the expression of antioxidant genes (such as HO1, NOQ1, and GCLC), which protects RPE, lens epithelial, and retinal ganglion cells from hydrogen peroxide-induced oxidative injury. In parallel, miR‐626 also modulates cell proliferation and apoptotic processes by inhibiting mTOR signaling through direct targeting of SLC7A5, suggesting a role in the pathogenesis of age-related macular degeneration (AMD). Notably, the dysregulation of miR‐626 is evident not only in ocular tissues but also in the cerebrospinal fluid—where its significantly reduced expression in Parkinson's disease (PD) patients points to its potential as a diagnostic biomarker."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of cancer biology, particularly in oral squamous cell carcinoma (OSCC), miR‐626 functions as an oncogenic modulator. Functional studies reveal that miR‐626 targets RASSF4, thereby promoting tumor cell growth. In addition, miR‐626 has been identified within tumor-derived exosomes, suggesting a role in mediating intercellular communication that facilitates tumor progression. Equally important, inhibition of miR‐626 in OSCC cells has been shown to suppress cell proliferation and enhance radiosensitivity—potentially through the downregulation of NF-κB signaling mediated by TRAF-interacting protein with forkhead-associated domain B—highlighting its promising therapeutic target value."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xiang-Zhong Xu, Yu Tang, Li-Bo Cheng, et al.
