MIR630 Gene

HGNC Family Non-coding RNAs
Name microRNA 630
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐630 (miR-630) has emerged as a pivotal regulator in diverse cancers, mediating distinct and context‐specific effects. In pancreatic cancer cells, overexpression of miR-630 induces apoptosis through downregulation of the insulin-like growth factor-1 receptor (IGF-1R), thereby promoting programmed cell death and suggesting a pro-apoptotic role ["}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "]. In gastric cancer, miR-630 is frequently upregulated; its elevated expression correlates with increased proliferation, invasion, metastasis, and poor patient survival, as also evidenced by its inverse relationship with circRNA_100269—a regulatory RNA that, when overexpressed, suppresses tumor cell growth ["}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "].\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond influencing cell survival, miR-630 exerts multifaceted control over proliferation and the apoptotic balance. It inhibits cell proliferation by targeting cell-cycle kinase 7 (CDC7), thereby blocking DNA synthesis and inducing G1 arrest, while simultaneously modulating several apoptotic regulators—including PARP3, DDIT4, and EP300—to maintain an equilibrium between cell death and survival under genotoxic stress ["}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "]. Moreover, under hypoxic conditions, miR-630 is upregulated and targets Dicer, a critical enzyme in microRNA biogenesis, leading to its downregulation. This reduction in Dicer levels contributes to decreased global miRNA expression, enhanced tumor growth, and metastasis, thereby highlighting an oncogenic aspect of miR-630 in the hypoxic tumor microenvironment ["}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "].\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a distinct context, hepatocellular carcinoma (HCC) studies reveal that miR-630 can function as a tumor suppressor. In HCC cells, overexpression of miR-630 attenuates the epithelial-mesenchymal transition (EMT) by directly targeting the transcription factor Slug, leading to reduced cell migration and invasiveness. Clinically, lower expression of miR-630 is associated with higher recurrence rates and shorter overall survival, underscoring its role as an inhibitor of metastatic progression in liver cancer ["}, {"type": "fg", "children": [{"type": "fg_f", "ref": "6"}]}, {"type": "t", "text": "].\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Lulu Farhana, Marcia I Dawson, Farhan Murshed, et al.
