| HGNC Family | Non-coding RNAs |
| Name | microRNA 634 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐634 (miR‑634) has emerged as a multifaceted tumor suppressor across a variety of malignancies. In esophageal, cervical, gastric, hepatocellular, nasopharyngeal, glioma, and tongue cancers, miR‑634 is frequently downregulated. Restoration or overexpression of miR‑634 induces apoptosis and reduces cell proliferation, migration, and invasion by targeting key oncogenic pathways. For example, enforced miR‑634 expression activates the mitochondrial apoptotic pathway via concurrent targeting of genes involved in mitochondrial homeostasis, antiapoptosis, antioxidant defense, and autophagy; it is also capable of suppressing the mTOR signaling pathway and downregulating targets such as JAG1, Rab1A, DHX33, and MAP2K4. Moreover, in several tumor types, competitive endogenous RNAs—including circular RNAs and long noncoding RNAs (e.g., DANCR and circRNAs)—sequester miR‑634, thereby derepressing oncogenic targets (such as GRB2) and contributing to chemoresistance and tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its tumor‐suppressive actions, miR‑634 plays important regulatory roles in non‐malignant conditions. In osteoarthritis, for instance, elevated miR‑634 levels in high‑grade chondrocytes suppress cell survival and matrix synthesis via direct targeting of PIK3R1, leading to reduced activation of the Akt/mTOR/S6 signaling axes. Similarly, in inflammatory vascular disorders such as ANCA‑associated vasculitis, increased miR‑634 in monocyte‑derived macrophages promotes a proinflammatory phenotype by enhancing ADAM17 and IL‑6 expression. Furthermore, in the context of hypertension, miR‑634 is downregulated in human aortic smooth muscle cells; its enforced expression inhibits Ang II–induced proliferation and migration by directly targeting Wnt4 and blocking β‑catenin nuclear translocation. Finally, the localization of miR‑634 within an intron of the PRKCA gene hints at its broader involvement in susceptibility to diverse inflammatory and neuropsychiatric conditions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "11", "end_ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xiaowei Peng, Peiguo Cao, Dong He, et al.
