MIR640 Gene

HGNC Family Non-coding RNAs
Name microRNA 640
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have demonstrated that miR‑640 plays an important regulatory role in angiogenesis. In vascular endothelial cells, exposure to hydrogen sulfide (H₂S) downregulates miR‑640 expression, thereby relieving its inhibitory effect on hypoxia inducible factor 1‑α (HIF1A) and facilitating the VEGFR2–mTOR signaling cascade that promotes angiogenesis. Similarly, treatment with angiopoietin‑1 (Ang‑1) decreases miR‑640 levels, which in turn upregulates the expression of Zinc Finger Protein 91 (ZFP91) and enhances endothelial cell migration and capillary‑like tube formation. These findings suggest that miR‑640 functions as a negative regulator of proangiogenic signaling in response to different vascular stimuli."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in angiogenesis, miR‑640 has been implicated in tumor biology. In renal cell carcinoma, a circular RNA (circ_0054537) acts as a competing endogenous RNA to sponge miR‑640, leading to upregulation of neuronal pentraxin‑2 (NPTX2) and promoting malignant progression. Moreover, in glioma cells, reduced miR‑640 activity is associated with increased expression of slit guidance ligand 1 (SLIT1) and activation of the Wnt/β‑catenin signaling pathway, which in turn decreases cellular radiosensitivity. Collectively, these studies highlight miR‑640 as an important tumor suppressor whose downregulation can contribute to enhanced tumor growth and therapy resistance."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yu Zhou, Xing-Hui Li, Cai-Cai Zhang, et al.
