| HGNC Family | Non-coding RNAs |
| Name | microRNA 641 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐641 is emerging as a multifunctional regulator implicated in several human diseases. In lung cancer, for example, miR‐641 is downregulated and its overexpression can inhibit cell proliferation and induce apoptosis via targeting MDM2 and upregulating p53, while in certain non–small‐cell lung cancer (NSCLC) contexts, increased miR‐641 expression—by directly targeting NF1 and activating ERK signaling—contributes to acquired resistance to EGFR‐targeted therapies. Furthermore, deregulation of the CRNDE/miR‐641/CDK6 axis in NSCLC supports its role as a negative regulator of cell growth and survival."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn other cancer types, miR‐641 functions in a context‐dependent manner. In cervical cancer, elevated levels of miR‐641 promote migration and invasion by modulating the TUSC8/PTEN axis. Likewise, in cholangiocarcinoma and bladder cancer, circular RNAs (CDR1as and circSETD3, respectively) sponge miR‐641, thereby attenuating its suppression of key oncogenic pathways such as AKT/mTOR or releasing inhibition of PTEN, which suggests an oncogenic contribution of endogenous miR‐641 in these settings. In breast cancer, studies have demonstrated that miR‐641 exerts tumor‐suppressive effects by targeting molecules such as SRCAP and RRS1; its activity is modulated by competing endogenous RNAs like LINC00665 and SNHG1, where sponging of miR‐641 reverses its growth‐inhibitory roles."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "9"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncogenesis, miR‐641 has been implicated in nonmalignant disorders.
