MIR648 Gene

HGNC Family Non-coding RNAs
Name microRNA 648
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nResearch has shown that miR-648 functions as a posttranscriptional regulator by targeting the 3′ untranslated region of endothelin-1 (ET-1) mRNA. In endothelial cells, elevated levels of placenta growth factor (PlGF)—a hallmark in sickle cell anemia patients with pulmonary hypertension—lead to a marked repression of miR-648. This downregulation of miR-648 results in an increased expression of the potent vasoconstrictor ET-1, thereby contributing to the development of pulmonary hypertension."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFurther investigation revealed that miR-648 is co‐transcribed with its host gene, MICAL3, from its distal promoter (P1), which is subject to positive regulation by the transcription factor PAX5. Downregulation of PAX5 activity by PlGF correlates with decreased miR-648 levels. Thus, the PlGF-induced reduction in PAX5-dependent transcription of miR-648 indirectly favors the stabilization and elevated levels of ET-1 mRNA, establishing a molecular link between microRNA regulation and vascular pathology in sickle cell anemia."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Chen Li, Caryn S Gonsalves, Marthe-Sandrine Eiymo Mwa Mpollo, et al.
