| HGNC Family | Non-coding RNAs |
| Name | microRNA 649 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nA growing body of literature has investigated key regulators of genomic stability, cellular differentiation, and development. Several studies have focused on components of the cohesin complex—most notably SMC3—which is essential for sister chromatid cohesion, proper mitotic and meiotic segregation, and the execution of complex transcriptional programs governing hematopoiesis, oocyte integrity, and even cardiac morphogenesis. Disruptions in SMC3 function have been linked to developmental anomalies, tumor suppressor dysregulation, and aberrant chromatin architecture in diverse biological contexts, ranging from lymphoma and myeloid malignancies to congenital heart disease and pancreatic cancer. These investigations underscore the central role of cohesin and its accessory pathways in maintaining genome integrity and proper cellular function."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast to these studies, miR-649 has been identified as a critical regulator within the viral replication framework. Specifically, research has demonstrated that miR-649 promotes herpes simplex virus type 1 (HSV-1) replication by directly targeting MALT1—a key mediator of the NF‑κB signaling pathway—without compromising host cell viability. Notably, miR-649 expression is downregulated following HSV-1 infection and is inversely correlated with MALT1 levels, suggesting that this microRNA plays a nuanced role in modulating antiviral signaling and may serve as a promising target for the development of novel antiviral therapies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Alexander Kurze, Katharine A Michie, Sarah E Dixon, et al.
