MIR6501 Gene

HGNC Family Non-coding RNAs
Name microRNA 6501
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nA review of the extensive literature provided reveals that many studies have focused on the multifaceted roles of transforming growth factor–β (TGF‐β) signaling and its downstream effectors—principally the Smad proteins—in regulating key cellular processes such as fibrosis, epithelial–mesenchymal transition, immune modulation, tumor progression, and development. These articles collectively dissect how TGF‐β signals via canonical (Smad2/3‐mediated) and noncanonical pathways to fine‐tune cellular outcomes through mechanisms that include differential phosphorylation, nuclear translocation, modulation by coactivators or corepressors, and cross-talk with other signaling networks such as MAPK, PI3K, and Wnt."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": "\n\nIn studies addressing pathological states, investigators have demonstrated that dysregulation of the TGF‐β/Smad axis is closely linked to disease progression in contexts such as chronic viral infections, fibrotic disorders, and advanced cancers. These works highlight that dynamic regulation—through processes such as Smad linker phosphorylation, adjustments in Smad protein turnover, and strategic interactions with other signaling molecules—can pivot the TGF‐β response from tumor suppression toward tumor promotion, and similarly, orchestrate the balance between tissue repair and fibrogenesis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "12"}]}, {"type": "t", "text": "\n\nOther investigations have probed the therapeutic potential of manipulating TGF‐β receptor activity or Smad function, demonstrating that targeted modulation of these pathways can attenuate fibrosis, limit metastatic behavior, and restore immune homeostasis. In various experimental models, intervention strategies that disrupt or enhance Smad signaling have led to marked changes in disease phenotypes, underscoring the clinical importance of this regulatory network."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "13", "end_ref": "18"}]}, {"type": "t", "text": "\n\nNotably, however, throughout this diverse body of work no study explicitly addresses or profiles the function of MIR6501. Although the regulatory complexity of the TGF‐β/Smad axis is explored in great depth—often in relation to cellular growth, differentiation, and stress responses—there is no direct evidence within these abstracts to indicate that MIR6501 plays a modulatory role in these processes. In the absence of any specific functional or mechanistic data regarding MIR6501 within the assembled literature, its biological role remains to be defined."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "25"}]}, {"type": "t", "text": "\n\nFinally, additional reports that delve into the transcriptional, post‐translational, and epigenetic regulation of cell fate decisions during development and tissue regeneration further elaborate on the sophisticated interplay of TGF‐β signals without implicating MIR6501 in these cascades.
