MIR6512 Gene

HGNC Family Non-coding RNAs
Name microRNA 6512
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nExcessive inflammation, as seen in periodontitis, impairs the osteogenic differentiation of human dental pulp stem cells (hDPSCs) through the upregulation of inflammatory cytokines such as TNF‑α. Under these inflammatory conditions, miR‑6512‑3p expression is increased, and this microRNA acts as a negative regulator of osteo/dentinogenic differentiation. In this context, the long noncoding RNA SNHG7, whose expression is downregulated by TNF‑α, normally functions as a molecular sponge that binds to miR‑6512‑3p, thereby reducing its abundance and mitigating its inhibitory effects on differentiation."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFunctionally, the suppression of miR‑6512‑3p by SNHG7 under inflammatory conditions alleviates the TNF‑α‑induced impairment of hDPSC differentiation. This is evidenced by improvements in key osteogenic markers, including enhanced calcium deposition, increased alkaline phosphatase activity, and elevated expression of differentiation markers such as BSP, DSPP, DMP‑1, RUNX2, and OPN. Thus, miR‑6512‑3p plays an inhibitory role in hDPSC osteogenic differentiation in an inflammatory microenvironment, a process that can be counteracted by SNHG7, offering potential therapeutic targets for the treatment of periodontitis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Wenyu Chen "}, {"type": "b", "children": [{"type": "t", "text": "SNHG7 promotes the osteo/dentinogenic differentiation ability of human dental pulp stem cells by interacting with hsa-miR-6512-3p in an inflammatory microenvironment."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Biochem Biophys Res Commun (2021)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1016/j.bbrc.2021.09.081"}], "href": "https://doi.org/10.1016/j.bbrc.2021.09.081"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "34653678"}], "href": "https://pubmed.ncbi.nlm.nih.gov/34653678"}]}]}]}
