MIR6746 Gene

HGNC Family Non-coding RNAs
Name microRNA 6746
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nCyclin D3 functions as a pivotal regulator of cell cycle progression by forming active complexes with CDK4/6 and ensuring proper G1‐to‐S phase transition in a variety of cell types. In lymphoid lineages, cyclin D3 is essential for the expansion of immature T cells and pre–B cells, where its regulated expression and timely degradation—mediated by specific kinases such as DYRK1A—help couple proliferation with cellular differentiation and immunoglobulin recombination. Dysregulation of cyclin D3 contributes to lymphoid malignancies and perturbs allelic exclusion of antigen receptor genes, thereby amplifying transformation risk."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its canonical cell cycle role in hematopoietic cells, cyclin D3 exerts important functions in nonhematopoietic tissues. In the uterus, cyclin D3 is critical for proper stromal cell decidualization—a process that involves both proliferation and subsequent polyploid differentiation—where its expression is modulated by hormonal and cytokine signals. In adipocytes, cyclin D3 acts in a cell cycle–independent manner by interacting with nuclear receptors to coactivate transcription and drive adipogenesis, while in the liver and muscle, distinct regulatory pathways govern its modest induction or stabilization, ensuring that proliferative cues are finely balanced with differentiation and metabolic programming."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, cyclin D3 participates in broader transcriptional and signaling networks that coordinate cellular responses to external stimuli. It contributes to the formation and expansion of germinal center B cells by modulating dark zone proliferation in response to T cell–dependent signals and interleukin‐21, and its aberrant levels can affect cellular transformation in fibroblasts and other lineages. Additional studies describe how cyclin D3 complexes with partner kinases control not only cell cycle progression but also the repression or activation of lineage‐specific gene programs, thereby influencing outcomes from skeletal muscle differentiation and regeneration to erythroid precursor divisions and cell fate decisions in B cells."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "11", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these studies underscore that cyclin D3 has multifaceted roles extending well beyond a simple cell cycle mediator—it integrates extracellular cues with intracellular responses to regulate proliferation, differentiation, and even genomic stability in a tissue‐ and context–dependent manner. Although the specific microRNA MIR6746 was not directly addressed in these reports, the extensive body of work on cyclin D3 suggests that any modulation by MIR6746 in similar systems could influence these critical processes. Future investigations into such microRNA–mediated regulation may further unravel how cyclin D3’s expression and activity are fine‐tuned in both normal physiology and disease."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "N Tiefenbrun, D Melamed, N Levy, et al.
