MIR6779 Gene

HGNC Family Non-coding RNAs
Name microRNA 6779
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Recent findings have revealed that the circRNA circSORBS1, which is notably underexpressed in lung cancer tissues, blood, and cell lines, functions by interacting with microRNAs. Specifically, circSORBS1 sequesters miR-6779-5p, thereby influencing downstream gene expression and contributing to the regulation of apoptosis and tumor behavior."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n\nDisruption of the normal interaction between miR-6779-5p and its target mRNA is a critical aspect of this regulatory pathway. Under typical conditions, miR-6779-5p would promote the degradation of RUFY3 mRNA; however, circSORBS1 binds miR-6779-5p as a molecular sponge. This sequestration indirectly prevents RUFY3 mRNA degradation, while direct binding between circSORBS1 and RUFY3 mRNA further enhances its stability, resulting in elevated levels of RUFY3 protein."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n\nThe increase in RUFY3 protein subsequently activates the YWHAE/BAD/BCL2 apoptotic signalling pathway, thereby inhibiting lung cancer cell proliferation and migration while promoting apoptosis. This dual regulatory mechanism underscores the potential therapeutic value of targeting the circSORBS1/miR-6779-5p axis in lung cancer, as disrupting this finely tuned interaction can alter tumor progression and improve disease outcomes."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Haotian Xu, Yue Zheng, Jiaxi Wu, et al.
