MIR7977 Gene

HGNC Family Non-coding RNAs
Name microRNA 7977
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‑7977 has been implicated in modulating the bone marrow microenvironment in myeloid neoplasms. In acute myeloid leukemia and myelodysplastic syndrome, leukemic cells secrete extracellular vesicles enriched in miR‑7977, which are taken up by bone marrow mesenchymal stem cells. This results in a direct reduction in poly(rC) binding protein 1 expression, causing an aberrant decrease in key hematopoietic growth factors such as Jagged‑1, stem cell factor, and angiopoietin‑1. Moreover, miR‑7977 also indirectly affects additional gene targets, perturbing normal hematopoietic supportive capacities and contributing to the replacement of healthy stem cells by neoplastic clones. These findings highlight a novel mechanism by which leukemic cells can alter their niche."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of lung adenocarcinoma, elevated levels of exosomal miR‑7977 in patient serum have been associated with advanced tumor stages. Functional in vitro assays demonstrate that miR‑7977 behaves as a tumor suppressor in lung cancer cells, where its inhibition enhances cell proliferation and invasion while decreasing apoptosis. These data suggest that, aside from its role in hematopoiesis, miR‑7977 may also serve as a promising diagnostic biomarker as well as a functional modulator in lung cancer pathogenesis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditionally, in epithelial ovarian cancer the expression of exosomal miR‑7977 is significantly reduced compared to healthy controls, with correlations noted to inflammatory indices such as the neutrophil‑lymphocyte ratio, underscoring its potential utility as a diagnostic and prognostic biomarker in this setting. Although a separate study investigating miRNA profiles in CD4⁺ T cells from autoimmune Addison’s disease did not focus specifically on miR‑7977 function, its documented dysregulation in disparate pathological contexts suggests that miR‑7977 may play wider roles in immune regulation and intercellular communication. Collectively, these studies indicate that miR‑7977 is a multifaceted microRNA with contexts ranging from the modulation of the bone marrow niche in myeloid neoplasms to tumor suppression and biomarker potential in solid cancers."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Hiroto Horiguchi, Masayoshi Kobune, Shohei Kikuchi, et al.
