| Name | microRNA 921 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In one investigation, hsa‐miR‐921 was shown to interact with the 3′‐untranslated region of the carbonyl reductase 1 (CBR1) gene, leading to a significant reduction in luciferase reporter activity, protein expression, and enzymatic function in cells carrying the major rs9024 G allele. This miRNA’s regulatory effect on CBR1 was found to be dependent on the genotype, as a differential impact was observed when comparing cells with the G allele to those with the A allele."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n \nIn a separate study focusing on lung cancer, hsa‐miR‐921 was identified as a negative regulator of glutathione peroxidase 3 (GPx3) in A549 cells. Here, the miRNA bound directly to the GPx3 3′‐UTR, resulting in an inhibition of GPx3 expression—a key antioxidant enzyme involved in reducing oxidative stress—which may have implications for tumorigenesis and chemotherapeutic response."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n\nCollectively, these findings demonstrate that hsa‐miR‐921 contributes to the fine-tuning of gene expression by targeting the 3′‐UTRs of distinct genes involved in enzymatic metabolism and oxidative stress regulation. Its ability to modulate CBR1 expression in a genotype-dependent manner as well as to suppress antioxidant defense proteins like GPx3 suggests that hsa‐miR‐921 may play an important role in pharmacological drug metabolism and cancer pathophysiology."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "James L Kalabus, Qiuying Cheng, Javier G Blanco "}, {"type": "b", "children": [{"type": "t", "text": "MicroRNAs differentially regulate carbonyl reductase 1 (CBR1) gene expression dependent on the allele status of the common polymorphic variant rs9024."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "PLoS One (2012)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1371/journal.pone.0048622"}], "href": "https://doi.org/10.1371/journal.pone.0048622"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "23133646"}], "href": "https://pubmed.ncbi.nlm.nih.gov/23133646"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Jang-Yeol Choi, Byung Chull An, In Jung Jung, et al.
