| HGNC Family | Non-coding RNAs |
| Name | microRNA 99a |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Reduced miR‑99a levels in tumor tissues have been closely associated with aggressive clinicopathological features, enhanced stem‐like properties and metastatic behavior, as well as poorer patient survival, highlighting its clinical relevance as a prognostic biomarker."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistically, miR‑99a exerts its tumor‐suppressive function by directly targeting multiple key regulators of cell growth, survival, and differentiation. Several studies have identified the mammalian target of rapamycin (mTOR) as a prominent direct target, with miR‑99a–mediated mTOR downregulation resulting in cell cycle arrest, inhibition of proliferation, and enhanced apoptosis. In addition, miR‑99a regulates other oncogenic pathways by suppressing transcripts such as IGF‑1 receptor, fibroblast growth factor receptor 3 (FGFR3), and NOX4, as well as by controlling epigenetic networks—for example, through its repression by long noncoding RNAs like ANRIL in gastric cancer. Together, these molecular interactions position miR‑99a as a central node in the regulation of critical signaling cascades including PI3K/Akt/mTOR and cell cycle regulators such as CDC25A and HOXA1."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nThe preclinical data further suggest that restoration of miR‑99a can re‐sensitize cancer cells to therapeutic interventions.
