MIR99B Gene

HGNC Family Non-coding RNAs
Name microRNA 99b
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‑99b, a member of the miR‑99 family, has emerged as a critical regulator of cancer cell behavior with context‑dependent functions. In several malignancies—including prostate, colorectal, gastric, cervical, hepatocellular, non‑small cell lung, and certain colorectal and breast cancers—deregulated miR‑99b expression modulates cell proliferation, migration, and invasion by targeting key signaling regulators such as mTOR, IGF‑1R, claudin‑11, and PPP2CA. In prostate and cervical cancers, for example, downregulation of miR‑99b relieves repression of mTOR and other chromatin remodelers, which correlates with enhanced tumor growth and aggressive clinical behavior; conversely, in certain esophageal and hepatocellular carcinomas, elevated miR‑99b levels have been associated with pro‑metastatic effects. Moreover, exosomal transfer of miR‑99b variants has been implicated in promoting drug resistance and metastatic phenotypes by modulating the AKT/mTOR axis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "12"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn non‑neoplastic contexts, miR‑99b also plays pivotal roles in tissue repair, cellular differentiation, and immune modulation. Its upregulation during wound healing and endothelial differentiation supports angiogenesis and the acquisition of endothelial cell markers, thereby facilitating vascular regeneration. In immune cells, miR‑99b (often co‑expressed with miR‑125a and let‑7e) fine‑tunes inflammatory responses by targeting components of the TLR and mTOR signaling pathways. Beyond these roles, miR‑99b influences musculoskeletal homeostasis—it promotes chondrocyte senescence in osteoarthritis by repressing MFG‑E8, reduces protein synthesis in muscle cells through RPTOR suppression, and impairs osteoblast proliferation via inhibition of FGFR3 in osteoporosis. Preliminary evidence also suggests a potential impact on neuroprotective responses in cerebral ischemia."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "13", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Dandan Sun, Yong Sun Lee, Ankit Malhotra, et al.
