| HGNC Family | Metallothioneins (MT) |
| Name | metallothionein 1A |
| Description | This gene is a member of the metallothionein family of genes. Proteins encoded by this gene family are low in molecular weight, are cysteine-rich, lack aromatic residues, and bind divalent heavy metal ions. The conserved cysteine residues co-ordinate metal ions using mercaptide linkages. These proteins act as anti-oxidants, protect against hydroxyl free radicals, are important in homeostatic control of metal in the cell, and play a role in detoxification of heavy metals. Disruption of two metallothionein genes in mouse resulted in defects in protection against heavy metals, oxidative stress, immune reactions, carcinogens, and displayed obesity. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMT1A is a member of the metallothionein family that functions primarily as a versatile, cysteine‐rich metal‐binding protein. Its well‐documented ability to sequester toxic metals—such as cadmium, copper, and bismuth—and to buffer essential metals like zinc underlies its role in heavy metal detoxification and cellular metal homeostasis. Detailed mechanistic studies have revealed that MT1A binds metals noncooperatively at early stages and then forms discrete metal–thiolate clusters through more cooperative pathways; these multiple binding modes allow it not only to serve as an intracellular store of loosely held zinc for donation to metalloenzymes but also to capture and neutralize harmful metal ions. Structural investigations of apo–MT1A and its sequential metallation with cadmium, zinc, copper, and even bismuth underscore its importance as both a dynamic sensor and buffer for metals."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "14"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMT1A also plays a significant role in the regulation of cellular metabolism and the modulation of disease susceptibility. Multiple population‐based genetic studies have demonstrated that single nucleotide polymorphisms in the MT1A gene can alter its ability to release zinc, thereby impacting redox homeostasis, inflammatory signaling, and insulin regulation. Associations between specific MT1A polymorphisms and conditions such as type 2 diabetes, cardiovascular complications, and renal dysfunction indicate that subtle variations in MT1A function can predispose individuals to metabolic derangements and influence the outcome of toxic exposures. These genetic variations may also affect MT1A’s protective antioxidant roles, thereby modulating both tissue injury and longevity under chronic inflammatory conditions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "22"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its metal‐binding and detoxification activities, MT1A is emerging as an important mediator of cellular signaling and tissue homeostasis. Its expression in a variety of tissues has been exploited as a diagnostic or prognostic biomarker in cancers and other stress‐related disorders, while epigenetic and transcriptional regulation of the MT1A gene—by factors such as PU.1 and p53—further refines its activity during immune cell differentiation, cell proliferation, and responses to oxidative stress. In addition, MT1A’s ability to participate in intercellular metal transfer, as well as its altered expression upon environmental exposures, supports its role as an integrator of metal‐related signals that ultimately influence tissue remodeling, inflammatory responses, and even steroid responsiveness."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "23", "end_ref": "41"}]}, {"type": "t", "text": ""}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "B A Masters, E J Kelly, C J Quaife, et al.
