OR11H2 Gene

HGNC Family G protein-coupled receptors
Name olfactory receptor, family 11, subfamily H, member 2
Description Olfactory receptors interact with odorant molecules in the nose, to initiate a neuronal response that triggers the perception of a smell. The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nTrace amine‐associated receptor 1 (TAAR1) is an intracellular G protein‐coupled receptor that plays a central role in modulating dopaminergic neurotransmission. Activation of TAAR1 by endogenous trace amines and amphetamine‐related substances alters the firing rate of dopamine neurons—particularly in the ventral tegmental area—and regulates downstream responses such as dopamine release and locomotor activity. TAAR1 also influences receptor signaling by interacting with D2 dopamine receptors to affect pathways like the AKT/GSK3 cascade and modulates inwardly rectifying potassium channels, thereby impacting the sensitivity to psychostimulants. These functions have been revealed through studies in knockout and transgenic mouse models that display altered behavioral responses and drug sensitivity."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nTAAR1’s intracellular signaling is mediated by coupling to distinct Gα‐subunits such as G_S and G_13, which in turn activate protein kinase A (PKA) and RhoA‐dependent pathways. Such compartmentalized signaling not only adjusts dopamine transporter trafficking and receptor responsiveness, but also affects glutamatergic neurotransmission in regions like the prefrontal cortex. These processes contribute to alterations in synaptic plasticity and cognitive function and determine the neurotoxic or neuroprotective outcomes in response to drugs like methamphetamine, amphetamine, and l-DOPA. Genetic and pharmacological manipulations have further uncovered TAAR1’s capacity to modulate autoreceptor dynamics and overall monoaminergic tone."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its primary role in monoaminergic regulation, TAAR1 is implicated in the broader modulation of neural network homeostasis and behavior. Its activity has been shown to influence stress-induced dendritic remodeling and cognitive deficits, as well as modulate responses in reward circuits that affect alcohol and drug consumption. Moreover, TAAR1 functions extend to interactions with receptors such as the D1 and μ-opioid receptors, and evidence suggests that TAAR1 may also participate in immunomodulatory processes in macrophages. These multifaceted actions underscore the therapeutic potential of TAAR1 agonists in treating neuropsychiatric disorders, addiction, and neurodegenerative conditions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "18", "end_ref": "23"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Daniel A Nelson, Melanie D Tolbert, Sam J Singh, et al.
